If some are picked before this mission starts, they will respawn upon starting the mission. Essentially, you cannot collect the required quantity prior to receiving the mission request.
For regular herbs, 3 in-game days was usually enough to get them to grow back. Some orchids will eventually stop spawning until you reach the part of the Duchesses mission. I never needed to wait for exotics to grow back but I assume it would be the same. You need to get rid of at least one orchid in your satchel for them to spawn again.
Yes, they respawn after 3 days I think. Your best bet is to just keep advancing the time in the game to about a week or so and then looking at all 10 locations again carefully.
